KASPar v4.0 SNP Genotyping System
The v4.0 formulation requires no change to how KASP is run in customers labs, indeed it canbe seen as a silent change. The change in the formulation results in more tightly clustered and better seperated data in a number of assays. In addition it offers better control of NTC's in assays potentially prone to primer dimer formation. The net result of this is increased pass rates of assays, higher call rates and lower failure rates, particularly when the genome of interest is complex due to size or ploidy.
Please note - KASP v3.0 will remain as a product for the foreseable future
but due to its inferior performance will eventually be phased out.

What do I need to run KASP v4.0 in my lab ?
There are two components required for a KASP v 4.0 Genotyping Reaction.
1.
The first is the KASPar reagent. This contains the universal fluorescent reporting dyes, the enzyme, buffer and dNTP's This comes as a kit containing:
- Pack leaflet
- KASPar reagent
2. The second is a SNP (or in/del) specific primer mix. There are two options available for this.
Please note that previously purchased KBD or KOD assays will work with either KASP v3.0 or KASP v4.0
- A KASP by Design assay
- A KASP on Demand assay
Validation
KASPar v4.0 was introduced into the KBioscience High Throughput Service Lab on the 1st March 2011 after a number of large pilot studies where both versions were run in parallel. The resulting increase in quality led to improved efficiency and decreased repeat rates.
Beta testing at Customer Facility
Conditions: Hexaploid wheat DNA derived from Seeds. Data was generated in 1536 well plates.
Production data produced from DNA from wheat seeds using KASP 3.0 and KASP 4.0 at a customers facility
